DAPI Staining Solution (1 mg/mL),IC-20017

Description  

DAPI (4¡ä,6-Diamidino-2-Phenylindole) is a blue-fluorescent DNA stain widely used in fluorescence microscopy and flow cytometry for nuclear staining. This solution is provided at a concentration of 1 mg/mL in ultrapure water, ensuring consistent performance and ease of use in various applications, including fixed and permeabilized cell preparations.  

Key Features  

- High Affinity for DNA: Binds strongly to AT-rich regions in double-stranded DNA.  

- Bright Blue Fluorescence: Exhibits maximum excitation:358 nm,emission:461 nm.  

- Cell Membrane Impermeant: Suitable for staining fixed cells or permeabilized live cells.  

- Ready-to-Use Solution: Pre-diluted to 1 mg/mL for convenience.  

- Compatibility: Can be used alongside other fluorescent probes (e.g., FITC, TRITC) for multiplex assays.  

Usage

Sample Preparation  

1. Fix and permeabilize cells (if required) using appropriate methods (e.g., methanol/acetone fixation or detergent permeabilization).  

2. Wash cells with PBS (pH 7.4) to remove residual fixative.  

Staining Procedure  

1. Dilute DAPI stock solution (1 mg/mL) to a working concentration of 0.1¨C1 µg/mL in PBS or mounting medium.  

2. Apply the diluted solution to samples and incubate for 5¨C15 minutes at room temperature (protected from light).  

3. Rinse gently with PBS to remove excess dye.  

4. Mount samples with an antifade mounting medium (if required) and visualize using a fluorescence microscope with a DAPI filter set.  

Recommended Dilution  

- For most applications: 1:1000 to 1:2000 dilution (final concentration 0.5¨C1 µg/mL).  

- Adjust based on cell type and experimental conditions.  

Precautions  

1. Toxicity: DAPI is a known mutagen. Handle with gloves, protective clothing, and eye protection.  

2. Light Sensitivity: Protect from light during storage and use to prevent photobleaching.  

3. Disposal: Dispose of waste according to local regulations for hazardous chemicals.  

4. Storage: Store at 2¨C8¡ãC in the original container. Do not freeze. Stable for 12 months when stored properly.  

5. Compatibility: Avoid contact with strong oxidizers or acids.
