Storage and Stability

Protocol (For Reference Only)

Important: Mix the loading buffer thoroughly before use to ensure uniform composition. Centrifuge briefly after thawing to collect contents at the bottom of the tube.

1. Sample Preparation: Mix protein sample with 5X SDS-PAGE Protein Loading Buffer at a ratio of 4:1 (sample:buffer) to achieve a final 1X concentration. For example, add 5 µL of 5X loading buffer to 20 µL of protein sample.

2. Denaturation: Heat the mixture at 95-100¡æ for 5-10 minutes to denature proteins.

3. Cooling and Loading: Cool the samples to room temperature, centrifuge briefly to collect condensation, and load directly onto SDS-PAGE gel.

4. Electrophoresis: Run the gel according to standard SDS-PAGE protocols until the dye front reaches the desired position.

 

Precautions

1. Vortex or shake the buffer before use, as components may precipitate during storage.

2. Heating time may be adjusted based on protein characteristics; membrane proteins or proteins with extensive disulfide bonds may require longer heating.

3. Avoid repeated freeze-thaw cycles for bulk packaging; aliquot if needed.

4. For certain membrane proteins or heat-sensitive proteins, incubation at 37¡ãC for 30 minutes is recommended. Published literature or technical documentation may also be consulted for specific protein sample preparation guidelines.
