Product Introduction

CCL20, also known as macrophage inflammatory protein-3 alpha (MIP-3¦Á) and liver activation regulated chemokine (LARC), is a small cytokine belonging to the CC chemokine family, located on chromosome 2 in the human genome. It is expressed at low levels in the thymus, testis, prostate, and intestine. CCL20 binds to and acts on the chemokine receptor CCR6, attracting lymphocytes and mild neutrophils. It induces intracellular calcium mobilization and mediates cancer, various autoimmune diseases, and antimicrobial effects. CCL20 has antimicrobial activity and is associated with inflammatory bowel disease, rheumatoid arthritis, psoriasis, other autoimmune diseases, and tumor diseases. It can induce Treg cell invasion into tumor tissue, recruit dendritic cells to promote immunosuppression, and induce endothelial cell proliferation and neointima formation. This recombinant human CCL20 (Ala27-Met96) is expressed by E. coli with a tag-free format.

Biological Activity:

1. Fully biologically active when compared to standard. The biological activity determined by a chemotaxis bioassay using human T-lymphocytes is in a concentration range of 10.0-50.0 ng/mL.

2. Measured by its ability to chemoattract THP-1 human acute monocytic leukemia cells. The ED50 for this effect is ¡Ü10.35 ng/mL, corresponding to a specific activity of ¡Ý9.662¡Á10⁴ U/mg.

3. Measured by its ability to chemoattract BaF3 mouse pro-B cells transfected with human CCR6. The ED50 for this effect is <2 ng/mL, corresponding to a specific activity of >5¡Á10⁵ U/mg.

Protocol (For Reference Only)

Important: Centrifuge the vial briefly before opening to bring the contents to the bottom. Reconstitute under aseptic conditions.

1. Reconstitution: It is not recommended to reconstitute to a concentration less than 100 µg/mL in ddH₂O. For long-term storage, it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

2. Aliquoting: Apportion stock solutions into working aliquots and store at ¡Ü -20¡ãC. Avoid repeated freeze-thaw cycles.

3. Chemotaxis Assay: The biological activity can be determined by chemotaxis bioassay using human T-lymphocytes (10.0-50.0 ng/mL), THP-1 cells (ED50 ¡Ü10.35 ng/mL), or BaF3 cells transfected with human CCR6 (ED50 <2 ng/mL).

4. Working Concentration: Optimal working concentrations should be determined empirically for each cell type and experimental system.
