Product Introduction

This product is low density lipoprotein (LDL) labeled with DiI (1,1''''-dioctadecyl-3,3,3'''',3'''' -tetramethylindocarbocyanine perchlorate), a lipophilic fluorescent carbocyanine dye. LDL is the major cholesterol-carrying lipoprotein in plasma, responsible for transporting cholesterol to peripheral tissues through receptor-mediated endocytosis via the LDL receptor (LDLR). DiI-LDL is widely used as a fluorescent probe for studying LDL receptor-mediated endocytosis, cellular cholesterol uptake, and LDL receptor expression and function. Upon binding to LDL receptors on the cell surface, DiI-LDL is internalized through clathrin-mediated endocytosis and delivered to lysosomes, where the DiI fluorescence can be visualized by fluorescence microscopy or quantified by flow cytometry. DiI-LDL is extensively utilized in atherosclerosis research, lipoprotein metabolism studies, and investigations of receptor-mediated endocytosis pathways. The DiI fluorophore exhibits orange-red fluorescence with excitation/emission maxima of approximately 549/565 nm.

Storage and Stability

Protocol (For Reference Only)

Important: Protect from light. Avoid repeated freeze-thaw cycles. Use aseptic technique for cell culture applications.

1. Working Solution Preparation: Dilute DiI-LDL in serum-free culture medium or PBS to the desired working concentration (typically 5-20 µg/mL).

2. Cell Treatment: Add DiI-LDL working solution to cultured cells. Incubate at 37¡ãC for 2-4 hours for uptake studies. For binding studies, incubate at 4¡ãC for 1-2 hours to prevent internalization.

3. Washing: Remove the DiI-LDL-containing medium. Wash cells 2-3 times with PBS to remove unbound DiI-LDL.

4. Detection: Visualize by fluorescence microscopy using a rhodamine/TRITC filter set, or quantify by flow cytometry in the PE channel.

5. Competition Studies: To confirm LDL receptor specificity, pre-incubate cells with excess unlabeled LDL (e.g., 10-50¡Á excess) before adding DiI-LDL.
