D-Galactose, 59-23-4,IC-03202897(99%)

Product Introduction

D-Galactose is a naturally occurring monosaccharide that can be used in animal models to establish subacute aging models in mice and cataract models in rats. Chronic D-galactose exposure induces neurodegeneration in mice by enhancing caspase-mediated apoptosis, inhibiting neurogenesis and neuronal migration, and increasing oxidative damage. In addition, D-galactose-induced toxicity in mice is a useful model for studying mechanisms of neurodegeneration and for evaluating neuroprotective drugs and agents. Oral administration of D-galactose causes cognitive impairment accompanied by oxidative damage in rats. Cognitive impairment is observed in the open field test at weeks 4 and 6 of D-galactose treatment, and spatial memory impairment is observed in the radial maze test after week 6. D-Galactose is a classic agent for subacute aging and cataract models. It alters the structure of proteins and peptides by reacting with free amine groups, leading to the accumulation of advanced glycation end (AGE) products during non-enzymatic glycation. AGEs contribute to age-related diseases. Rats and mice are commonly used as animal models.

Product Features

1. Classic Aging Model Agent: Widely used to establish subacute aging models in mice and rats.

2. Cataract Model Agent: Used to induce cataract models in rodents.

3. Well-Characterized Mechanism: Induces AGE accumulation through non-enzymatic glycation and increases oxidative damage.

4. Neurodegeneration Research Tool: Chronic exposure induces neurodegeneration, cognitive impairment, and spatial memory deficits.

5. High Purity: Supplied at high purity for reproducible animal model studies.

Protocol (For Reference Only)

Important: Prepare solutions fresh before use. For in vivo administration, filter sterilize solutions before injection.

1. Solution Preparation: Dissolve D-galactose in sterile saline or PBS with sonication assistance (<60ˇăC) to prepare the desired concentration. Filter sterilize through a 0.22 µm membrane for in vivo use.

2. Subacute Aging Model (Mouse): Use female or male C57BL/6J mice (8 weeks old). Administer D-galactose subcutaneously at doses of 50, 100, or 200 mg/kg/day for 8 weeks.

3. Cataract Model (Mouse): Administer D-galactose subcutaneously at 200 mg/kg/day for 8 weeks.

4. Subacute Aging Model (Rat): Use albino-Wistar rats (120-130 g). Administer D-galactose intraperitoneally at 300 mg/mL/kg for 7 days.

5. Cataract Model (Rat): Administer D-galactose intraperitoneally at 15 g/kg twice daily for 30 days.

6. Assessment: Evaluate cognitive function using behavioral tests such as the open field test and radial maze test. Assess oxidative damage by measuring oxidative stress markers. Evaluate neurodegeneration by histological analysis.

Precautions

1. Prepare solutions fresh before use and filter sterilize for in vivo administration.

2. The optimal dose and administration route should be validated for specific animal strains and experimental objectives.

3. Monitor animal body weight and health status throughout the treatment period.

4. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: What is the mechanism of D-galactose-induced aging?

A1: D-galactose reacts with free amine groups on proteins and peptides, leading to the accumulation of advanced glycation end (AGE) products through non-enzymatic glycation, which contributes to age-related diseases. It also increases oxidative damage and induces caspase-mediated apoptosis.

Q2: What doses are used for subacute aging models in mice?

A2: D-galactose is typically administered subcutaneously at 50, 100, or 200 mg/kg/day for 8 weeks in C57BL/6J mice.

Q3: How should I store D-galactose?

A3: Store powder at -20ˇăC for up to 3 years or at 4ˇăC for up to 2 years. For solutions, store at -80ˇăC for up to 6 months or at -20ˇăC for up to 1 month.

Q4: What is the solubility of D-galactose?

A4: D-galactose is soluble in water at 62.5 mg/mL (with sonication assistance <60ˇăC) and in DMSO at 50 mg/mL.
