2¡Á Fast Taq PCR Master Mix, IC-5209

Product Introduction

2¡Á Fast Taq PCR Master Mix is an optimized 2¡Á concentrated PCR premix formulated with a modified Taq DNA Polymerase that exhibits an amplification speed approximately 4 times faster than conventional Taq DNA Polymerase, reaching up to 15 sec/kb. The formulation is further supplemented with a single-stranded DNA-binding protein, PCR enhancers, and protein stabilizers that work synergistically to improve amplification efficiency and sensitivity. It can amplify fragments up to 4 kb from complex genomic DNA or up to 5 kb from lambda DNA. The product is convenient to use: simply add 0.5 reaction volume of 2¡Á Fast Taq PCR Master Mix, primers, and template, and adjust the final volume with ddH₂O. The amplified products carry a single adenine (A) overhang at the 3'''''''' end, allowing direct cloning into T-Vectors. The mix contains a blue electrophoresis tracking dye, enabling direct agarose gel electrophoresis analysis of amplification products without additional loading buffer.

Product Features

1. Fast Amplification: Modified Taq DNA Polymerase enables amplification speeds up to 15 sec/kb, approximately 4 times faster than conventional Taq.

2. Enhanced Efficiency and Sensitivity: Single-stranded DNA-binding protein, PCR enhancers, and protein stabilizers work synergistically to improve amplification performance.

3. Long Fragment Amplification: Capable of amplifying fragments up to 4 kb from complex genomic DNA and up to 5 kb from lambda DNA.

4. Convenient 2¡Á Premix Format: Simply add primers and template, then adjust volume with ddH₂O.

5. Direct T-Vector Cloning: Amplified products carry a 3'''''''' A overhang for direct cloning into T-Vectors.

6. Direct Gel Loading: Contains blue electrophoresis tracking dye for direct agarose gel electrophoresis.

Protocol (For Reference Only)

Important: Thaw the Master Mix on ice and mix gently before use. Avoid repeated freeze-thaw cycles.

1. Reaction Setup: Prepare the PCR reaction on ice according to the following typical 50 µL reaction: 25 µL of 2¡Á Fast Taq PCR Master Mix, forward primer (final concentration 0.2-0.5 µM), reverse primer (final concentration 0.2-0.5 µM), template DNA (10-100 ng genomic DNA or 1-10 ng plasmid DNA), and ddH₂O to a final volume of 50 µL.

2. PCR Cycling: Use the following typical cycling conditions: initial denaturation at 94-95¡ãC for 1-3 minutes; 25-35 cycles of denaturation at 94-95¡ãC for 10-15 seconds, annealing at 50-65¡ãC for 15-30 seconds (optimize according to primer Tm), extension at 72¡ãC for 15 seconds per kb; final extension at 72¡ãC for 5 minutes.

3. Analysis: Load the PCR products directly onto an agarose gel for electrophoresis; the blue tracking dye allows direct loading without additional loading buffer.

4. Cloning (Optional): Purify the amplified product and clone directly into a T-Vector according to standard protocols.

Precautions

1. Thaw the Master Mix on ice and mix gently before use. Avoid vortexing.

2. Avoid repeated freeze-thaw cycles; aliquot into single-use volumes if needed.

3. Optimize annealing temperature and cycle number according to primer Tm and template complexity.

4. For GC-rich templates, consider adding additional enhancers or optimizing cycling conditions.

5. For research use only. Not for use in diagnostic or therapeutic procedures.
