Dialyzed Fetal Bovine Serum (FBS),IC-1912

Source: Premium Uruguayan Fetal Bovine Serum

Sterility: 0.1 µm membrane filtered

Preservation: Store at -10¡æ to -40¡æ. Avoid repeated freeze-thaw cycles.

Description

Dialyzed Fetal Bovine Serum is processed from premium-quality Uruguayan-sourced FBS. The serum undergoes an extensive dialysis procedure to remove low molecular weight molecules (typically below 10,000 Daltons), including glucose, hormones (e.g., insulin, thyroid hormones), vitamins, amino acids, and salts. This makes it an essential supplement for research applications where the precise control of these components is critical, such as in metabolic studies, hormone-responsive cell culture, and isotope-labeling experiments.

Uruguayan herds are recognized for their high health status and are free from major bovine diseases (as per OIE standards), ensuring a safe and consistent raw material source.

Key Features & Benefits

- Low Molecular Weight Depletion: Effectively removes molecules <10 kDa, reducing background interference in sensitive assays.

- Defined Basal Conditions: Provides a more controlled environment for studying the effects of specific nutrients, drugs, or signaling molecules.

- Ideal for Isotope Studies: Essential for metabolic tracer experiments using labeled glucose, amino acids, or nucleotides.

- Consistent Performance: Sourced from a single, traceable geographical region (Uruguay) to minimize batch-to-batch variability.

- Rigorous Safety Profile: The source serum is collected from approved abattoirs, and the final product is extensively filtered to ensure sterility and mycoplasma absence.

Typical Dialysis Specifications

Cut-off Membrane: 10 kDa nominal molecular weight cut-off (MWCO).

Removed Components :

Glucose: Typically reduced to <1 mg/dL.

Insulin: >95% removed.

Thyroid hormones (T3/T4): >95% removed.

Steroid hormones: Significantly reduced.

Most amino acids and vitamins: Significantly reduced.

Retained Components: High molecular weight growth factors, proteins, peptides, and other essential macromolecules necessary for cell growth and attachment are preserved.

Applications

- Studies of glucose metabolism, insulin signaling, or other hormone-sensitive pathways.

- Cell culture requiring the addition of defined concentrations of specific small molecules (e.g., inducible expression systems).

- Metabolic flux analysis and stable isotope-resolved metabolomics.

- Selection media for transfected cells (when used with specific lacking components).

- General cell culture where a reduced-hormone background is desired.

Quality Control

- Sterility Test: Negative for bacterial and fungal growth.

- Mycoplasma Test: Negative by PCR and/or culture.

- Endotoxin Level: Typically <10 EU/mL (Lot-specific certificate provided).

- Hemoglobin Content: <25 mg/dL (indicative of low hemolysis during collection).

- Total Protein: 30 - 45 mg/mL (consistent with high-quality FBS).

- Growth Promotion Test: Performed on standard cell lines (e.g., HeLa, CHO-K1, or Balb/3T3) to ensure performance efficacy.

- Viral Safety: Sourced from countries with negligible BSE risk (Uruguay is OIE recognized BSE risk-negligible country). Raw serum is screened for relevant bovine viruses.

Usage Instructions

1.  Thawing: Thaw slowly at 2-8¡æ overnight or in a water bath at 37¡æ with gentle swirling. Do not thaw at room temperature.

2.  Heat-Inactivation (Optional): If required, heat at 56¡æ for 30 minutes in a water bath with periodic gentle mixing. Rapid cooling is recommended post-inactivation. Note: Heat-inactivation may precipitate some proteins; clarification by centrifugation may be necessary.

3.  Supplementation: Because dialyzed FBS lacks essential low molecular weight nutrients, the basal culture medium MUST be supplemented accordingly (e.g., with glucose, glutamine, specific amino acids, or a custom supplement mix) to support cell growth. Optimization for your specific cell type is recommended.

4.  Recommended Concentration: Commonly used at 5-20% (v/v) in culture medium, depending on cell type and application.

5.  Storage: For long-term storage, keep at -10¡æ to -40¡æ. Aliquot into single-use volumes to avoid repeated freeze-thaw cycles, which can degrade product quality.

Precautions & Disclaimer
