Product Introduction
This product is a sterile, colloidal silica-based density gradient medium designed for the isolation and purification of cells, organelles, and viruses. Percoll consists of silica particles (15-30 nm diameter) coated with polyvinylpyrrolidone (PVP), which provides a non-toxic, low-osmolality, and low-viscosity medium suitable for the separation of viable cells and subcellular components.
Percoll is widely used for the isolation of blood cells (lymphocytes, monocytes, granulocytes), subcellular organelles (mitochondria, lysosomes), and viruses. It can be used either as a pre-formed continuous or discontinuous gradient, or mixed with cells to form a self-generating gradient upon centrifugation. The low endotoxin content and ready-to-use sterile formulation make it ideal for sensitive primary cell isolation and subsequent cell culture applications.

Protocol (For Reference Only)
Important: All steps should be performed under aseptic conditions. Undiluted Percoll should be adjusted to physiological osmolality before use by mixing with 1.5 M NaCl or 10X concentrated medium (9 parts Percoll : 1 part 1.5 M NaCl).
1. Gradient Preparation: Dilute Percoll to the desired density using 0.15 M NaCl or cell culture medium; for continuous gradients, centrifuge at 15,000-30,000 x g for 15-30 minutes.
2. Sample Loading: Carefully layer the cell suspension onto the prepared Percoll gradient.
3. Centrifugation: Centrifuge at 200-800 x g for 15-30 minutes at 4¡ãC using a swing-out rotor without brake.
4. Cell Collection: Harvest the purified cell layer from the appropriate density interface using a sterile pipette or syringe.
5. Washing: Dilute the collected cells with 3-5 volumes of PBS or medium, centrifuge at 300-400 x g for 5-10 minutes, and repeat wash twice to remove residual silica particles.
Precautions
1. Do not freeze. Freezing causes irreversible aggregation of the silica particles.
2. Adjust osmolality before use to prevent cell lysis or shrinkage.
3. Use a swing-out rotor for best band resolution.
4. Turn off the centrifuge brake to avoid disrupting the gradient interface.
