Red Blood Cell Lysis Buffer, IC-1187
Product Introduction
This product is a ready-to-use ammonium chloride-based buffer designed for the selective lysis of red blood cells (erythrocytes) in whole blood, bone marrow, or splenocyte suspensions. The hypotonic ammonium chloride solution rapidly disrupts the erythrocyte membrane while preserving the integrity and viability of nucleated white blood cells (leukocytes), making it an essential reagent for flow cytometry sample preparation, immunophenotyping, and primary cell isolation.
The buffer is formulated with optimized concentrations of ammonium chloride, potassium bicarbonate, and EDTA to ensure efficient lysis with minimal damage to leukocyte membranes. It is supplied as a sterile-filtered, ready-to-use 1X solution, eliminating the need for dilution or additional preparation steps.
Product Features
1. Ready-to-Use 1X Formulation: No dilution required, saving preparation time and reducing variability.
2. Gentle on Leukocytes: Optimized formula selectively lyses erythrocytes while preserving white blood cell viability and surface antigen expression.
3. High Efficiency: Complete lysis of red blood cells typically achieved within 5-10 minutes at room temperature.
4. Low Endotoxin: Sterile-filtered and tested for endotoxins, suitable for sensitive downstream applications including cell culture and functional assays.
5. Versatile Application: Compatible with whole blood, bone marrow, spleen homogenates, and tissue-derived single-cell suspensions.
Specifications
Size: Available in 100 mL and 500 mL
Storage and Stability
Storage Conditions: Store at 4¡ãC. Protect from light.
Shelf Life: The product is stable for 24 months from the date of manufacture when stored as directed.
Protocol (For Reference Only)
Important: Use cold lysis buffer for optimal results. Keep cells on ice after lysis to minimize leukocyte damage.
1. Sample Preparation: Collect whole blood or bone marrow in anticoagulant tubes (EDTA or heparin recommended) and mix gently.
2. Lysis: Add 10-20 volumes of 1X Red Blood Cell Lysis Buffer to the sample (e.g., 1 mL blood + 10-20 mL buffer), mix by gentle inversion, and incubate on ice or at room temperature for 5-10 minutes with occasional mixing until the suspension becomes clear red.
3. Centrifugation: Centrifuge at 300-500 x g for 5 minutes at 4¡ãC, carefully aspirate and discard the red supernatant without disturbing the white cell pellet.
4. Optional Repeat Lysis: If the pellet appears red, repeat steps 2-3 for incomplete lysis samples.
5. Wash and Resuspend: Resuspend the white cell pellet in PBS or desired buffer, wash once by centrifugation, and resuspend for downstream applications including flow cytometry staining, cell culture, or nucleic acid extraction.
Precautions
1. Avoid over-lysis as prolonged exposure may damage leukocyte membranes and reduce viability.
2. Use cold buffer (4¡ãC) for optimal preservation of leukocyte surface markers.
3. For samples with high nucleated red blood cell content (e.g., fetal liver, some leukemias), additional optimization may be required.
4. Do not freeze the buffer as repeated freeze-thaw cycles may alter buffer performance.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
FAQ (Simplified)
Q1: My white blood cell pellet still appears red after lysis. What should I do?
A1: Repeat the lysis step with fresh buffer or increase the buffer-to-sample ratio. Some samples with high red blood cell content may require two rounds of lysis.
Q2: Does this buffer affect cell surface markers for flow cytometry?
A2: No. The ammonium chloride-based formula is gentle and preserves most surface antigen expression. However, for sensitive epitopes, keeping the buffer and incubation time cold is recommended.
Q3: Can I use this buffer on tissue samples?
A3: Yes. It can be used on single-cell suspensions prepared from spleen, thymus, or lymph nodes to remove contaminating red blood cells.
Q4: What is the difference between 1X and 10X formulations?
A4: This product is supplied as a ready-to-use 1X solution. No dilution with distilled water is necessary, ensuring consistent osmolality and performance.
