Protocol (For Reference Only)

Important: Use aseptic technique when handling the sterile solution. Prepare 1X working solution freshly for optimal coating performance.

1. Preparation of 1X Working Solution: Dilute the 10X Poly-L-Lysine Solution 1:10 with sterile tissue culture grade water or sterile PBS to obtain a 0.01% (w/v) 1X working solution. Mix gently by swirling or inversion.

2. Surface Coating of Culture Vessels: Add sufficient 1X working solution to completely cover the culture surface (typically 0.5 mL per well of a 24-well plate, 1-2 mL per well of a 6-well plate, or 3-5 mL per T25 flask). Incubate at room temperature for 1 hour or at 37¡ãC for 30 minutes.

3. Aspiration and Washing: Aspirate the Poly-L-Lysine solution completely and rinse the coated surface 2-3 times with sterile tissue culture grade water or PBS to remove excess unbound PLL.

4. Drying: Allow the coated surface to air dry completely in a sterile biosafety cabinet with the lid slightly ajar for 1-2 hours. For urgent use, drying can be accelerated by brief exposure under UV light in the biosafety cabinet.

5. Cell Seeding: Once dry, seed cells onto the coated surface in appropriate culture medium and incubate under standard culture conditions. Coated plates may be used immediately or stored at 4¡ãC for up to one week wrapped in sterile packaging.

Precautions

1. Do not freeze the solution as freezing may cause polymer aggregation and loss of coating efficiency.

2. Aspirate the coating solution completely and wash thoroughly to remove excess unbound PLL, as residual free PLL in the culture medium may be cytotoxic to certain cell types.

3. The optimal coating concentration and incubation time may vary for different cell types; optimization is recommended for specific experimental systems.
