Rhodamine B Isothiocyanate, IC-01250637

Product Introduction

Rhodamine B isothiocyanate (RBITC) is a fluorescent dye commonly used in biological imaging and labeling applications. It contains an isothiocyanate functional group that can be covalently attached to amino groups on proteins and other biomolecules, making it useful for fluorescent labeling of cells and tissues. RBITC fluoresces bright orange-red under green light excitation, which facilitates detection and tracking of labeled molecules in complex samples. Due to its stability and sensitivity, RBITC has been widely used in various research fields, including cell biology, immunology, and neurobiology. In vitro, Rhodamine B isothiocyanate is a biochemical reagent that can be used as a biological material or organic compound for life science research.

Product Features

1. Amine-Reactive Labeling: Isothiocyanate group covalently reacts with primary amines on proteins and biomolecules.

2. Bright Orange-Red Fluorescence: Excitable by green light, facilitating detection in complex samples.

3. Good Stability and Sensitivity: Suitable for long-term imaging and tracking applications.

4. Broad Research Utility: Widely used in cell biology, immunology, and neurobiology.

5. High Purity: Supplied at 95% purity for reproducible labeling.

Shelf Life: The product is stable for 24 months from the date of manufacture when stored as directed in powder form.

Protocol (For Reference Only)

Important: RBITC is moisture-sensitive and light-sensitive. Prepare solutions freshly before use and protect from light. Use anhydrous solvents for stock solutions.

1. Stock Solution Preparation: Dissolve RBITC in anhydrous DMSO or DMF to prepare a concentrated stock solution (e.g., 10-50 mM). Prepare fresh before use and protect from light.

2. Protein Labeling: Dilute the protein to be labeled in 0.1 M sodium carbonate/bicarbonate buffer, pH 9.0-9.5, at a concentration of 1-10 mg/mL. Add RBITC stock solution at a molar ratio of 5-50:1 (RBITC:protein). Incubate at room temperature for 1-2 hours or at 4¡ãC overnight with gentle mixing, protected from light.

3. Purification: Remove unreacted RBITC by gel filtration chromatography, dialysis, or ultrafiltration. Determine the degree of labeling by measuring absorbance at the appropriate wavelength.

4. Storage of Conjugates: Store purified conjugates at 4¡ãC protected from light in the presence of a preservative for short-term use, or aliquot and store at -20¡ãC for long-term storage. Avoid repeated freeze-thaw cycles.

5. Working Dilution: Determine the optimal working dilution empirically for each application.

Precautions

1. Protect from light and moisture during handling and storage.

2. Prepare stock solutions in anhydrous solvents and use freshly.

3. Conjugation efficiency is affected by buffer pH, protein concentration, and molar ratio; optimization is recommended.

4. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: What is the reactive group of RBITC?

A1: RBITC contains an isothiocyanate group that reacts with primary amines on proteins and other biomolecules to form stable thiourea linkages.

Q2: What buffer should I use for protein labeling?

A2: Use 0.1 M sodium carbonate/bicarbonate buffer at pH 9.0-9.5. Avoid buffers containing primary amines such as Tris or glycine.

Q3: How should I store RBITC?

A3: Store powder at 4¡ãC sealed, away from moisture and light. For solutions, store at -80¡ãC for 6 months or -20¡ãC for 1 month (sealed, away from moisture and light).
